翼飞雪总RNA快速提取试剂可以在1小时的时间,从动物组织、植物组织、培养细胞、细菌细胞等样本中提取完整的总RNA,提取的总RNA A260/A280值约在1.6~1.8之间。另外,提取过程中加入氯仿后,保留下层的酚相可提取DNA和蛋白质。

本总RNA快速提取试剂可以从1mg组织样本中提取约1-10μg 总RNA,从106培养细胞中提取5-15μg 总RNA,由于本试剂提取的总RNA可以很好的避免DNA或者蛋白质的污染,因此提取产物总RNA可以用于Poly(A+)筛选、体外翻译、RNA印迹分析、斑点杂交、cDNA文库构建、RT-PCR、荧光定量PCR等后续分子生物学实验

产品特点

1、产品中的RNApure,为总RNA提取试剂中的氯仿替代物,其使用效果与氯仿完全相同,但是由于其会发点高达120℃以上,因此常温不挥发,避免了氯仿常温挥发对人体带来的损害。

样本处理方式

1、组织液氮研磨:取适当组织放入研钵中,加入液氮迅速研磨,待组织变软后 继续加入 液氮研磨,反复三次后,按照 1mL总RNA快速提取试剂:50-100mg组织的比例加入提取 试剂,进行后续提取操作。

2、组织匀浆:按照 1mL 总 RNA 快速提取试剂:50-100mg 组织的比例加入提取试剂,用 电动匀浆机充分匀浆 1-2min,进行后续提取操作。 注:裂解产物应呈澄清透明装粘稠液体,如组织为肌肉、脂肪、植物结节部位等富含蛋白、脂肪、多糖的样本,匀浆后可能仍 会残留有部分不溶物质,可 4℃ 12,000g 离心 10min,取上清至于零一洁净离心管中。

3、贴壁培养细胞:按照 1mL 总 RNA 快速提取试剂:10cm2培养细胞的比例加入提取试剂, 直接进行消化裂解,进行后续提取操作。

4、悬浮培养细胞:动物细胞、植物细胞、酵母细胞按照 1mL 总 RNA 快速提取试剂:5×106细胞;细菌细胞按照 1mL 总 RNA 快速提取试剂:5×107细胞的比例加入提取试剂, 直接收集、裂解,进行后续提取操作。

注意事项

1、 RNA 的提取质量,很大程度上决定于样品的选择以及保存方式,因此建议尽 可能使用新鲜的样品提取 RNA。如果样品采集后不能立即进行 RNA 提取,应将新采集的新鲜样本放入液氮 中速冻,然后置于-80℃长期保存。

2、样品采集后立即浸泡在本试剂盒中的样本裂解液中,4℃条件下可保存 1 周。

3、理论上,裂解液体积:样品量的比值越大,提取的 RNA 质量越好。本说明书中的比例仅为推荐比例,最佳使用比例还需用户自行摸索。我们建议不清楚样品中 RNA 含量时, 开始时使用较少的样品进行提取,然后再根据实验具体情况调整样品量。

4、仔细阅读本说明书,每个品牌的产品都有各自特性,切忌按照使用其他品牌 试剂盒的提取经验,来使用本试剂。

5、塑料制品处理:尽可能使用一次性无菌塑料制品,已标明 RNase-Free 的塑料制品, 未开封前可直接使用。未标明 RNase-Free 的塑料制品处理方式:在玻璃杯中倒 入去离子水,加入 DEPC 至其终浓度为 0.1% (DEPC 活性很强的剧毒物,需在通 风橱中小心使用)。将待处理的塑料制品放入一个可高温高压灭菌的容器中,倒 入配制好的 DEPC 水,使塑料制品的所有部分都浸泡在溶液中,在通风橱中 37℃ 或室温下处理过夜。将 DEPC 水小心倒入废液瓶中,将处理过的塑料制品连同容 器以铝箔封口,高温高压灭菌 30min,然后将塑料制品在 80℃烘烤干燥,置于洁净处备用。

6、玻璃和金属制品处理:250℃烘烤 3 小时以上。

7、RNase 另一污染源是移液器,根据移液器制造商的要求对移液器进行处理。 一般情况采用 DEPC 配制的 70%乙醇擦洗移液器的外部和内部,基本达到要求。
