翼飞雪 CCK-8(Cell Counting Kit)试剂盒是一种基于 WST-8 的广泛应用于细胞增殖 和细胞毒性的快速灵敏试剂盒。

WST-8 是一种类似于 MTT 的化合物,在电子耦合试剂 1-Methoxy PMS 存在的情况下, 可被还原生成橙黄色水溶性甲臜(Formazan),细胞增殖越快越多,甲臜溶液颜色越深,细 胞毒性越大,甲臜溶液颜色越浅。对于同样的细胞,颜色的深浅和细胞的数目呈线性关系。

翼飞雪 CCK-8 检测试剂盒可适用于绝大多数细胞系和实验环境。

注意事项

1.如果待测物质有氧化性或者还原性,可以在加入 CCK-8 试剂之前更换新鲜培养基(除去 培养基,并用新鲜培养基洗涤细胞两次,然后加入新鲜培养基),去掉药物影响。如药物影 响比较小,可以不更换培养基,直接扣除培养基中加入药物后的空白吸收即可。

2.建议先做几个孔摸索接种细胞的数量和加入 CCK-8 试剂后的培养时间。

3.有条件时建议使用多通道移液器,可以有效减少平行孔间的差异。加入 CCK-8 试剂时, 建议斜贴培养板壁加入,而不要查到培养基下面加入,以避免产生气泡,干扰 OD 值读数。

4.白细胞可能需要培养较长时间。

5.当使用标准 96 孔板时,贴壁细胞的最小接种量至少为 1,000 个细胞/孔(100µL 培养基)。 由于白细胞灵敏度相对较低,因此检测白细胞时最小接种量至少为 2,500 个细胞/孔(100µL 培养基)。如要使用 24 孔板或 6 孔板实验,应先计算每孔相应的接种量,并按照每孔培养 基总体积的 10%加入 CCK-8 试剂。

6.如果没有 450nm 的滤光片,可以使用吸光度在 430-490nm 之间的滤光片,但是 450nm 滤光片的检测灵敏度最高。

7.培养基中酚红的吸光度可以在计算时,通过扣除空白孔中本底的吸光度而消除,因此不 会对检测结果造成影响
